Thinking that once the whole quilt is done, you won't be able to notice.. Truly think we are our worse judges! Looks nice!
I totally agree with the galloping horse rule and the 3 yard rule....you need to remember that we sit right on top of what we sew, it seems to magnify every mistake...standing back gives you a better perspective! And a more accurate one as well!
I don't think you will ever be happy with the quilt unless you fix it. While I agree that no one will notice it, you always will. It would not take much time to fix that one block.

I would fix it but that's just me. Depends on how much it bothers you. If you're not a perfectionist then let it go. The fact that you're asking suggests to me that you don't want to fix it. If that's the case, just leave it as it is. I see two places where they don't line up. The most noticeable to me is bottom left block. The fact that it is only 12 blocks makes it more noticeable. They wouldn't be that noticeable on a much larger quilt.

Looks fine to me. I tend to be overly critical of my own work but I find no one else is as critical as I am. The recipient will just be delighted to be getting a quilt made with love.
If you need more convincing; old quilting wisdom is that every quilt should contain some flaw because "only God is perfect".

Quilting is something that we enjoy doing. If we get to the point that we only focus on our mistakes, then we won't enjoy it. I try to do as little ripping out as possible. Once in awhile, I have something that I can't live with, but I've found that a lot of things that were an issue when I was piecing together don't look as bad later on when it is completely quilted and the binding is on.
The top is complete. And I have to sit and look to find that one mistake.
